Interest in functional mushrooms as potential supports for stress and anxiety has grown considerably in recent years, reflecting broader recognition that certain bioactive compounds found in fungi may interact with neurological and endocrine systems relevant to psychological wellbeing. The research base for this application is uneven across species, with some mushrooms showing human clinical data and others representing mechanistic or preclinical evidence only. This review organizes the current state of evidence by species, examines proposed mechanisms, and identifies where findings are strong, preliminary, or still largely theoretical.
The Biology of Stress and Why Mushrooms Are Being Studied
The stress response is coordinated primarily by the hypothalamic-pituitary-adrenal (HPA) axis, which regulates cortisol secretion, and the autonomic nervous system, which governs the sympathetic “fight or flight” response. Chronic activation of these systems is associated with a range of adverse health outcomes, including mood disturbance, sleep disruption, impaired immune function, and increased risk of metabolic disease.
Several functional mushrooms contain bioactive compounds that may interact with components of this system. The specific mechanisms proposed include: stimulation of neurotrophic factors such as nerve growth factor (NGF) and brain-derived neurotrophic factor (BDNF), which support neuronal plasticity and stress resilience; modulation of the HPA axis through adaptogenic triterpenoid compounds; interaction with GABAergic and serotonergic signaling pathways; and anti-inflammatory activity that may reduce neuroinflammation associated with chronic stress.
None of these mechanisms has been demonstrated as definitively causally effective in humans in the way that pharmaceutical interventions have been. What the research provides is a growing set of converging signals that particular species may offer modest, biologically plausible contributions to stress physiology. The distinction between a plausible mechanism and a clinically confirmed effect is important to maintain throughout this review.
Lion’s Mane (Hericium erinaceus): The Most Studied Species for Mood
Hericium erinaceus has received more focused investigation for mood-related and stress-adjacent outcomes than any other functional mushroom. Its unique bioactive compounds, hericenones (from the fruiting body) and erinacines (from the mycelium), stimulate NGF and BDNF synthesis in preclinical models. Both NGF and BDNF are proteins involved in neuronal survival, synaptic plasticity, and the regulation of mood-relevant circuits, particularly in the hippocampus and prefrontal cortex.
Human Trial Evidence
A randomized, double-blind, placebo-controlled pilot study published in Nutrients examined the effects of H. erinaceus supplementation in 41 healthy young adults aged 18 to 45. Participants in the active group received 1.8 g of Lion’s Mane daily for 28 days. The trial found that participants in the active group performed significantly faster on the Stroop task at 60 minutes post-dose, suggesting an acute effect on cognitive processing speed. After 28 days of supplementation, a trend toward reduced subjective stress was observed, though this did not reach conventional statistical significance (p = 0.051). The authors noted the small sample size as a limitation and called for larger trials to confirm the finding.[1]
Mechanistic Context
A review published in Antioxidants examined the potential antidepressant mechanisms of H. erinaceus alongside other botanical compounds. The authors identified several biochemical pathways by which Lion’s Mane may exert psychotropic activity: rescue of neurotransmitter imbalances associated with depression and anxiety, stimulation of neurogenesis through NGF and BDNF pathways, counteraction of oxidative stress and mitochondrial dysfunction, and attenuation of neuroinflammation. The review described this constellation of effects as broadly mimicking the mechanisms of conventional antidepressants, though with a notably different and generally favorable tolerability profile. The authors also emphasized that further human trials are needed before clinical conclusions can be drawn.[2]
The Lion’s Mane research is the most mature in the functional mushroom space for stress and mood outcomes. Even so, the available human data remains limited in scale. The mechanistic rationale is strong and the preclinical work is consistent, but large confirmatory RCTs have not yet been published.
Reishi (Ganoderma lucidum): Adaptogenic and Fatigue-Related Evidence
Ganoderma lucidum has a long history of use in traditional Chinese medicine as a tonic for vitality, stress resilience, and nervous system support. Its two primary bioactive compound classes, polysaccharides and triterpenoids (particularly ganoderic acids), have been studied through distinct mechanistic lenses relevant to stress physiology.
The triterpenoids are of particular interest for their proposed adaptogenic properties. In preclinical models, ganoderic acid fractions have demonstrated sedative-hypnotic activity, inhibition of certain inflammatory signaling pathways, and potential modulation of HPA axis activity. The polysaccharide fraction, meanwhile, has demonstrated immunomodulatory activity that may indirectly influence stress-related outcomes via the gut-brain axis and neuroimmune signaling.
Clinical Evidence for Fatigue and Wellbeing
While direct clinical trials examining Reishi specifically for anxiety disorder are limited, a notable randomized, double-blind, placebo-controlled study published in the Journal of Medicinal Food investigated a Ganoderma polysaccharide extract in 132 patients with neurasthenia, a condition characterized by chronic fatigue, mood disturbance, and diminished stress tolerance. Patients received a Ganoderma polysaccharide preparation or placebo for 8 weeks. At the end of the study, the treatment group showed significantly lower fatigue scores (reduction of 28.3% versus 20.1% in placebo) and a greater increase in sense of wellbeing (38.7% improvement versus 29.7% in placebo). The global clinical improvement was also significantly different between groups. The authors concluded that the Ganoderma preparation was superior to placebo for symptom improvement in this population.[3]
While neurasthenia is not equivalent to an anxiety disorder, the symptom overlap is clinically meaningful. Fatigue, emotional exhaustion, and impaired stress coping are features shared by both conditions. This trial represents the most robust human clinical evidence currently available for Reishi’s effects on stress-adjacent outcomes.
Other Functional Mushrooms With Relevant Preclinical Evidence
Cordyceps
Cordyceps militaris is most studied for energy metabolism and exercise capacity, but its primary bioactive compound cordycepin (a nucleoside analog) has demonstrated neurological activity in preclinical research. Cordycepin modulates adenosine receptor signaling and cyclic AMP (cAMP) pathways that intersect with stress-response neurotransmitter systems. Some animal studies have reported reductions in anxiety-like behavior following Cordyceps extract administration, but no dedicated human anxiety trials have been published. The evidence at this stage is mechanistic and preclinical.
Chaga (Inonotus obliquus)
Chaga’s dense polyphenol, melanin, and triterpenoid content positions it as one of the most antioxidant-rich functional mushrooms. Chronic oxidative stress contributes to dysregulation of stress response systems, and preclinical work has examined Chaga extracts in models of anxiety and neurotoxicity. While the antioxidant rationale is scientifically credible, direct human data examining Chaga for stress or anxiety outcomes does not currently exist in the peer-reviewed literature.
Reishi Spores and Fruiting Body Comparisons
Some research has begun distinguishing between the stress-relevant activity of Reishi’s fruiting body, mycelium, and spore preparations, as the triterpenoid and polysaccharide profiles can vary considerably between these forms. Spore oils tend to be richer in triterpenoids, while fruiting body preparations tend to contain higher beta-glucan content. For the proposed adaptogenic and sedative mechanisms most relevant to stress, dual-extract preparations capturing both fractions may be more relevant, though this comparison has not been formally evaluated in stress-specific clinical trials.
Mechanisms Shared Across Species
Across the species reviewed, several recurring mechanisms appear relevant to stress and anxiety outcomes:
- Neurotrophic factor stimulation: Lion’s Mane erinacines and hericenones stimulate NGF and BDNF, proteins involved in neuronal resilience and stress adaptation
- Neuroinflammation modulation: Multiple species, including Reishi and Chaga, contain compounds that inhibit pro-inflammatory cytokines relevant to the neuroimmune stress response
- HPA axis interaction: Reishi triterpenoids have demonstrated preclinical activity on cortisol-related pathways, though human HPA axis data is limited
- Gut-brain axis signaling: Polysaccharides from multiple species function as prebiotic substrates and may influence serotonin and GABA precursor availability through microbiota-mediated pathways
This overlap in mechanisms suggests that multi-species combinations may offer complementary, rather than redundant, support for stress physiology. Practical Considerations
Several factors are worth bearing in mind for anyone evaluating functional mushrooms for stress support:
- Evidence hierarchy: Lion’s Mane has the most direct human data for stress-adjacent outcomes. Reishi has a relevant clinical trial for fatigue and wellbeing. Other species currently have preclinical evidence only for anxiety and stress-specific outcomes.
- Product quality: The bioactive content of functional mushroom supplements varies widely by extraction method, source material, and whether fruiting body, mycelium, or both are included. Supplement labels do not consistently specify the compounds most relevant to stress mechanisms (such as erinacines or ganoderic acid concentrations).
- Interaction awareness: Reishi in particular has documented potential for interaction with medications affecting immune function, platelet aggregation, and blood glucose. Individuals taking prescription medications for anxiety, depression, or stress-related conditions should consult a healthcare provider before adding any functional mushroom supplement to their routine.
- Adjunctive framing: No functional mushroom has evidence supporting its use as a standalone treatment for clinical anxiety disorder. The research positions these species as potential complementary supports for general stress resilience, not replacements for evidence-based therapeutic interventions.
Summary
Of the functional mushrooms currently studied for stress and anxiety-related outcomes, Lion’s Mane has the most specific and developed research base, including a human pilot RCT showing a trend toward reduced subjective stress after 28 days. Reishi has a human clinical trial demonstrating improvements in fatigue, wellbeing, and clinical global impression in neurasthenia patients. Other species including Cordyceps and Chaga carry mechanistically relevant but primarily preclinical evidence. Across all species, the research is promising but not yet sufficient to support definitive clinical claims. The biological rationale is credible, the tolerability profile is generally favorable, and the field continues to develop.
