
Hericium erinaceus (Lion’s Mane) and Ganoderma lucidum (Reishi) are two of the most extensively studied functional mushrooms in the scientific literature. Both have long histories of use in East Asian traditional medicine and both have been the subject of published human clinical trials. However, their primary bioactive compounds, proposed mechanisms, and areas of strongest research support differ considerably. This comparison outlines what the current evidence shows for each species and offers a framework for thinking about which may be more relevant depending on individual goals.
Bioactive Profiles: Where the Species Diverge
Understanding the differences between Lion’s Mane and Reishi begins with their distinct compound profiles.
Lion’s Mane is unique among functional mushrooms for containing two classes of compounds not found in other species: hericenones, which are present in the fruiting body, and erinacines, which are found in the mycelium. Both compound families have been identified as stimulators of nerve growth factor (NGF) synthesis in preclinical research. NGF is a protein that supports the growth, maintenance, and survival of neurons. Lion’s Mane also contains beta-glucan polysaccharides with immunomodulatory properties, but the neurotrophic compounds are its most distinctive bioactive feature.
Reishi is characterized by a complex mixture of triterpenes (particularly ganoderic acids), polysaccharides including beta-glucans, and fungal immunomodulatory proteins (FIPs). The triterpenes, which require alcohol extraction to isolate, have been studied for anti-inflammatory, antioxidant, and immunomodulatory effects. The polysaccharide fraction of Reishi has been examined for immune support. Reishi does not contain hericenones or erinacines and does not share Lion’s Mane’s proposed neurotrophic mechanism.
Lion’s Mane: Cognitive Function and Neurological Support
The most robust clinical evidence for Lion’s Mane centers on cognitive function and mood. A landmark double-blind, placebo-controlled trial published in Phytotherapy Research examined oral administration of H. erinaceus fruiting body in 30 adults aged 50 to 80 who had been diagnosed with mild cognitive impairment. At weeks 8, 12, and 16, participants in the Lion’s Mane group showed significantly higher scores on a cognitive function scale compared with the placebo group. Scores increased progressively with continued intake, and at four weeks after discontinuation, cognitive scores decreased significantly, suggesting the effects were tied to ongoing supplementation.[1]
A later randomized, double-blind, placebo-controlled parallel-groups study conducted at Northumbria University examined both acute and chronic effects of H. erinaceus in 41 healthy young adults aged 18 to 45. After a single dose, participants performed significantly faster on the Stroop task, a measure of cognitive processing speed and inhibition, at 60 minutes post-dose. A trend toward reduced subjective stress was observed after 28 days of supplementation, though the authors noted the sample size was small and results should be interpreted cautiously.[2]
A further randomized crossover study examined a single dose of Nordic-grown Lion’s Mane extract in healthy adults. At two hours post-ingestion, Lion’s Mane was associated with significant improvements in working memory performance, complex attention, and reaction time, as well as subjective ratings of happiness, compared with placebo.[3]
The NGF Hypothesis
The proposed mechanism underlying these cognitive effects involves stimulation of NGF synthesis by hericenones and erinacines. NGF supports the maintenance and repair of cholinergic neurons in the basal forebrain, which are involved in memory and attention. This pathway is biologically plausible and well-characterized in cell culture and animal models. Demonstrating this mechanism directly in human brain tissue remains a challenge, but the clinical outcomes observed in trials are consistent with the hypothesis.
Reishi: Immune Modulation and Fatigue
Reishi’s research base is oriented primarily around immune function, fatigue, and inflammatory markers, rather than direct cognitive support. A pilot randomized clinical trial published in Evidence-Based Complementary and Alternative Medicine examined spore powder of G. lucidum in 48 breast cancer patients undergoing endocrine therapy who reported cancer-related fatigue. The intervention group showed statistically significant improvements in fatigue and physical wellbeing scores after four weeks, as well as reductions in anxiety, depression, and improved quality of life. Notably, circulating concentrations of TNF-alpha and IL-6, both pro-inflammatory cytokines associated with fatigue and depressive symptoms, decreased significantly in the treatment group, with no serious adverse effects reported.[4]
A more recent study published in Journal of Advanced Research examined Ganoderma lucidum spore powder as a supplementary intervention alongside COVID-19 vaccine boosters in human participants. Using single-cell multi-omics analysis, researchers found that GLSP significantly enhanced neutralizing antibody responses in participants with moderate baseline immunity. At the cellular level, GLSP expanded classical monocyte populations, strengthened IFN-alpha responses, and promoted broader antiviral immune activity. The active compound identified as driving the IFN-alpha response was the triterpenoid ganoderic acid H.[5]
Reishi and Sleep: A Secondary Area of Research
Reishi has also been investigated for potential effects on sleep quality, with some animal research suggesting that polysaccharide fractions may influence sleep duration and architecture. Human clinical evidence for sleep-specific effects remains limited, and this area requires further investigation before firm conclusions can be drawn. Individuals interested in mushrooms specifically for sleep support may find that Reishi is included in some combination formulations for this purpose, but the evidence base is less developed than for immune-related outcomes.

The two mushrooms occupy different positions in the functional mushroom research landscape:

Choosing Between Them: A Research-Based Framework
There is no universally superior choice between Lion’s Mane and Reishi. The more relevant question is what biological outcome is most important to the individual, and which species has more direct supporting evidence for that outcome.
For individuals primarily interested in cognitive support, processing speed, or neurological resilience, Lion’s Mane is the species with more targeted clinical evidence. Its neurotrophic mechanism is biologically distinct from other functional mushrooms, and human trials have demonstrated cognitive outcomes in both impaired and healthy populations.
For individuals whose primary interest is immune support, management of fatigue, or reduction of inflammatory markers, Reishi’s evidence base is more directly applicable. Its triterpene fraction and polysaccharides have been studied in human populations in contexts involving immune activation and inflammatory modulation.
Some individuals may use both species in combination. There is no established evidence of adverse interaction between them, and their mechanisms are sufficiently distinct that they are unlikely to be redundant. Supplement Quality Considerations
For both species, the source material and extraction method influence what active compounds are present in the final product.
For Lion’s Mane, fruiting body extracts are the source of hericenones; erinacines are found in the mycelium. Some research has used whole fruiting body powder rather than extract. For maximum transparency, products should specify their source material and, where possible, the extraction method used.
For Reishi, a dual extract (combining hot water extraction for polysaccharides and alcohol extraction for triterpenes) is generally considered more comprehensive than a single water extract alone. Products listing both beta-glucan and triterpene content provide more meaningful quality information than those listing only total polysaccharide content.
Safety
Both Lion’s Mane and Reishi have been used in human clinical trials without reports of significant adverse effects. A pharmacovigilance review of adaptogenic and immunomodulatory natural products noted that Ganoderma lucidum appeared among products with some serious adverse event reports in a WHO database, though causality was difficult to establish due to multi-ingredient products and incomplete reporting, and the overall safety profile in controlled trials has been favorable.[6] Individuals with autoimmune conditions, those taking anticoagulant medications, or those who are pregnant or breastfeeding should consult a healthcare provider before using either species in supplement form.
